Yaohai Novagen has announced another major achievement. Following the successful launch of its first recombinant humanized type III collagen, which addressed the high sensitization risk associated with conventional collagen products, the company has now achieved another breakthrough: the first successful expression of recombinant humanized type III collagen with a high hydroxylation rate using a prokaryotic expression system.
This breakthrough marks another important step in Yaohai Novagen’s use of synthetic biology to advance the medical-aesthetics industry.

Why Hydroxylation Matters for Collagen
The structural stability and biological activity of collagen are closely related to its degree of hydroxylation. In natural collagen, proline hydroxylation is a key step in forming a stable triple-helix structure and directly affects thermal stability, solubility, and cellular affinity.
Collagen with insufficient hydroxylation often struggles to form a stable triple helix and may have limited biological activity. Achieving a high hydroxylation rate has therefore remained one of the central challenges in recombinant collagen development and is an important indicator of product quality and function.

Hydroxylation Rate: 35%
The hydroxylated recombinant humanized type III collagen developed by Yaohai Novagen achieved a proline hydroxylation rate of approximately 35%, approaching the natural level of animal collagen, which is about 37%.
Circular Dichroism Analysis
A characteristic positive signal for the triple-helix structure appeared near 220 nm, clearly confirming that the product possesses a complete tertiary conformation and a highly ordered helical structure.

Transmission Electron Microscopy
The sample displayed distinct fiber-bundle morphology, a typical ultrastructural feature of natural collagen. This further confirmed the strong biomimetic structural characteristics and self-assembly capability of the hydroxylated collagen.

The successful development of highly hydroxylated humanized collagen represents another major milestone for Yaohai Novagen following its advances in humanized collagen, recombinant PDRN, and recombinant botulinum toxin.
